摘要
目的探讨在妊娠期高血压疾病(HDCP)发病中白血病抑制因子(LIF)对围产儿结局的影响。方法分别采用酶联免疫吸附试验检测37例子痫前期和50例妊娠期高血压孕妇外周血LIF水平,荧光定量RT-PCR(FQ-RT-PCR)技术检测胎盘组织LIF-mRNA的表达情况,以80例正常足月妊娠妇女作为对照,并详细记录围产儿的情况。结果 (1)子痫前期组孕妇外周血LIF水平低于妊娠期高血压组和正常妊娠组孕妇,差异有统计学意义(P<0.05),而后两组之间差异无显著性。(2)子痫前期组LIF-mRNA的表达较妊娠期高血压组和正常对照组明显降低,差异有统计学意义(P<0.01),且随着病情的加重,LIF-mRNA表达下降。妊娠期高血压组和正常妊娠组比较,LIF-mRNA的表达稍有降低,但无统计学差异(P>0.05)。(3)孕妇外周血LIF水平与新生儿评分有关,与新生儿出生体重(r=0.47;P<0.05)和胎盘重量(r=0.36;P<0.05)均存在显著性正相关;与胎儿胎龄无明显相关。结论 HDCP孕妇血清LIF水平降低,胎盘组织中LIF-mRNA表达明显下降,LIF可能对围产儿的结局有一定影响。
Objective To study the effect of leukemia inhibitory factor(LIF) on the prognosis of the neonates delivered by womem with hypertensi on of pregnancy.Methods 37 women with preeclampsia,50 women with gestational hypertension and 80 normal pregnant women were studied.The levels of LIF in maternal venous serum and the expression of LIF in the placenta tissue from normal pregnant women and those with preeclampsia and gestational hypertension were detected by enzyme linked immunosorbent(ELISA) and quantitative fluorescent reverse transcription-polymermal chain reaction(QF-RT-PCR) technique respectively.The outcome of neonates were recorded carefully.Results(1)The levels of LIF in maternal venous serum with preeclampsia were significantly lower than those with gestational hypertension and normal pregnancies(P0.05),although there was no substantial difference between gestational hypertension and normal pregnancies.(2)There were statisatically significant decreases of LIFmRNA from preeclampsia and gestational hypertension and normal pregnancies(P0.01),but no statistical difference between gestational hypertension and normal pregnancies(P0.05).(3)There was correlations between the levels of LIF in maternal venous serum and neonate apgar score.By Pearson correlation analysis,there was positive correlation between the LIF concentration and newborn birth weight(r=0.47;P0.05) and placental weight(r=0.36;P0.05).Conclusion The serum levels of LIF and the expression of LIFmRNA in placental tissues were significantly reduced in the women with preeclampsia.LIF might in part detrimentally affect the prognosis of the neonates delivered by women with hypertension of pregnancy.
