三氧化二砷对人宫颈癌HeLa细胞凋亡及其血管内皮生长因子表达的影响 被引量:4

摘要 目的了解三氧化二砷(As2O3)对人宫颈癌HeLa细胞凋亡及其血管内皮生长因子(VEGF)表达的影响。方法用不同浓度的As2O3作用于HeLa细胞不同时间段后,光镜和电镜观察细胞形态改变,MTT法测定细胞生长抑制情况,流式细胞术检测细胞凋亡率,Western blotting方法检测VEGF的表达。结果不同浓度的As2O3处理HeLa细胞不同时间后,能抑制宫颈癌Hela细胞的体外生长,诱导宫颈癌Hela细胞凋亡,并呈浓度和时间依赖性(P<0.01)。As2O3还下调VEGF的表达。结论 As2O3可抑制体外人宫颈癌HeLa细胞增殖及VEGF的表达。 Objective To study the effect of arsenic trioxide (As2O3) on apoptosis and VEGF expression of human HeLa cells. Methods HeLa cells were treated with As2O3 at various concentrations for different time points, and their growth inhibi-tion, apoptosis and VEGF expression were determined by MTT, flow cytometry and Western blotting, respectively. Results As2O3 pretreatment induced the growth inhibition and apoptosis of HeLa cells in a dose-and time-dependent manner (P0.01), and downregulation of VEGF expression. Conclusion As2O3 can in vitro inhibit cell proliferation and VEGF expression of human HeLa cells.
