摘要
采用SDS-酶裂解法提取观音土曲的细菌DNA。提取的DNA进行16S rDNA的PCR扩增、克隆测序并构建进化树,结果表明获得的三个有效克隆分别属于未培养细菌和胚芽乳杆菌。探讨了采用分子生物学技术鉴定观音土曲中功能细菌的可行性。
The bacterial DNA ofGuanyin Tuqu was extracted by SDS alkaline lysis method. The DNA extracts were employed for the PCR-amplification of 16S rDNA. The PCR products were cloned and sequenced. Then the phylogenetic tree was constructed. The obtained three clones belonged to uncultured bacteria and Lactobacillus plantarum, respectively. The probability of identifying functional bacteria in Guanyin Tuqu by molecular techniques was indicated by this study.
