摘要
目的 观察乙肝病毒X蛋白(HBx)对甲硫氨酸腺苷转移酶1A(MAT1A)、甲硫氨酸腺苷转移酶2A(MAT2A)表达的影响.方法 应用免疫组织化学法检测78例乙肝病毒(HBV)阳性肝细胞癌(HCC)组织及其癌旁组织中HBx和MAT1A、MAT2A的表达,分析其阳性表达,并分析它们对HCC临床病理参数的影响.结果在78例HCC组织中,HBx、MAT1A、MAT2A阳性表达分别为59例(75.64%)、17例(21.79%)和66例(84.62%);HBx表达和MAT1A表达呈负相关(P〈0.05),和MAT2A表达呈正相关(P〈0.05);MAT1A和MAT2A表达呈负相关(P〈0.05).HBx阳性表达和TNM分期、病理分级、血清甲胎蛋白(AFP)正相关;MAT1A阳性表达和肝硬化程度、血清AFP含量呈负相关;MAT2A阳性表达和肝硬化程度、肿瘤大小、TNM分期呈正相关.结论 肝癌组织中HBx蛋白和MAT1A、MAT2A的表达明显相关,在HCC发生、发展过程中它可能通过影响MAT1A、MAT2A而实现MATs表达的转变.
Objective To investigate possible effect of hepatitis B virus x-protein (HBx) on the expression of methionine adenosyltransferase-1A ( MAT1A ) and methionine adenosyltransferase-2A ( MAT2A),and to elucidate possible mechanism of MATs transformation in the process of oncogenesis and development of hepatocellular carcinoma (HCC).Methods Exam the expression of HBx,MAT1 A,MAT2A in 78 HBV-positive ( HCC tissues and the adjacent tissues of tumor by IHC method,compare the expressive relationships among them,and analyze the expressions of them with some clinicopathological parameters.Results In 78 carcinoma specimens,the positive expressions of HBx,MAT1 A,MAT2A were 59 cases (75.64% ),17 cases (21.79%) and 66 cases(84.62% ) ;the relationship between the expressions of HBx and MAT1A was negative (P 〈0.05 ),between HBx and MAT2A was positive (P 〈0.01 );between MAT1A and MAT2A was negative (P 〈 0.05 ).The positive expression of HBx,MAT1A,MAT2A were 55 cases (70.51% ),25 cases (32.05% )and 57 cases (73.08%)in 78 cases of adjacent tissues of tumor.The expression of HBx has positive relationship to TNM stages,pathological grades and serum alpha fetoprotein (AFP).The expression of MAT1A has negative relationship to liver cirrhosis and serum AFP.The expression of MAT2A has positive relationship to liver cirrhosis,tumor sizes and serum AFP.Conclusion The expression of HBx has relationship to the expressions MAT1A,MAT2A in HCC tissues,maybe Hbx can regulates the expression of MATs by inhibiting MAT1 A and inducing MAT2A;the expressions of HBx,MAT1A and MAT2A were relate to some important clinicopathological parameters.
