摘要
为研究板蓝根生物碱酸性部位(RIAa)抗新城疫病毒(NDV)的机制,通过细胞病变抑制法和MTT比色法检测了RIAa对NDV感染鸡胚成纤维细胞(CEF)吸附和释放的影响。结果显示:药毒混合方式中当RIAa质量浓度为31.3μg·mL-1、作用60min时A570值显著高于病毒对照(P<0.05),在120min时3个浓度均显著高于病毒对照(P<0.05);先加药物方式中RIAa为62.5和31.3μg·mL-1在120min时均显著高于病毒对照(P<0.05);先加毒吸附方式中,各吸附时间的A570值与病毒对照差异均不显著(P>0.05)。RIAa+NDV的细胞上清液再次作用细胞后其A570值显著高于没有加RIAa的NDV对照(P<0.05)。空斑试验结果表明:RIAa高、中、低3个药物浓度作用孔的空斑数都显著低于病毒对照(P<0.05),以中质量浓度(7.8μg·mL-1)的空斑减少率最高,进一步证实了RIAa的抗NDV作用。结论:吸附时间、加药方式对病毒感染细胞的能力有显著影响,RIAa在一定浓度下对病毒的吸附有阻碍作用,能够保护细胞减少病毒感染,但是不能解除已经吸附的病毒;RIAa在一定浓度下能够显著抑制病毒的释放。
Effect of acid of radix isatidis alkaloid ( RIAa) on adsorption and release of Newcastle disease virus ( NDV) was detected by cytopathic effect inhibition and MTT colorimetric assays. The results showed that the A570 value of RIAa at the concentration of 31. 3 μg·mL -1 when absorbed 60 min in simultaneous adding RIAa and virus after mixed was significantly higher than that of virus control ( P 〈0. 05). However,the A570 values of RIAa at the three concentrations when absorbed 120 min were significantly higher than that of virus control ( P 〈0. 05). A570 values of RAIa at the concentrations of 62. 5 and 31. 3 μg·mL -1 when ab-sorbed 120 min in pre-adding drug were significantly higher than that of virus control ( P〈 0. 05). However,A570 values in postadding drug of different absorbed time showed no significant difference with that of virus control ( P 〉0. 05). When adding cell supernatant into chick embryo fibroblast ( CEF) again,the A570 values of RIAa + NDV were significantly higher than that of virus control ( P〈 0. 05). Plaques numbers treated with different concentrations of RIAa were significantly lower than that of virus control. Antivirus ability of RIAa was confirmed by the results of plaque formation assay. Conclusion:absorption time and the methods of adding drugs showed significant effect on the ability of virus infecting cells. RIAa at a certain concentration could inhibit virus absorbing on cells,and protect cells from infecting virus. However,it could not remove virus which had absorbed on cells. On the other hand,RIAa could remarkablely inhibit virus's releasing.
