摘要
目的:建立维药刺糖中黄酮类化合物含量测定的荧光分析法。方法:根据黄酮类化合物的荧光性质,以芦丁为标准品,用荧光分析法测定了刺糖中黄酮类化合物的含量;并系统考察了溶剂、pH值、放置时间对荧光强度的影响。结果:在70%的乙醇中加入pH值7.0的三酸缓冲溶液,芦丁荧光强度最大,线性范围在2.7×10^-7-1.35×10^-5mol·L^-1,r=0.999 5,平均回收率为98.9%(RSD=1.9%,n=9)。结论:该方法准确度高、灵敏度高,适用于药用刺糖中黄酮类化合物的含量测定。
Objective:To establish fluorescence method for the determination of flavone contents in saccharum alhagi.Methods:According to fluorescent character of flavonoids,the flavone contents in saccharum alhagi were determined by spectrofluorimetry.Results:The fluorescence was the strongest while solvent was 70% ethanol with Britton-Robinson buffer(pH 7.0).The linear concentration range of rutin was 2.7×10^-7-1.35×10^-5mol·L^-1 with a correlation coefficient of 0.9995.The average recovery of samples was 98.9%(n=9,RSD=1.9%).Conclusion:The method is simple,rapid and accurate,which is suitable for determination of flavone contents in saccharum alhagi.
