摘要
目的:构建肝细胞生长因子cDNA真核表达载体。方法:提取大鼠胎肝总RNA,利用RT-PCR技术扩增出肝细胞生长因子cDNA,连接于载体pMD18-T质粒上,构建重组载体,经双酶切和DNA测序分析检测重组载体构建结果。结果:成功构建肝细胞生长因子cDNA真核表达载体。结论:为进一步研究HGF与肝癌的关系提供了实验基础。
Objective:To construct expression vector of HGF cDNA. Methods: We extracted the fetal liver total RNA, used RTPCR to amplify HGF cDNA, and then cloned them in pMD18 -T vector to construct recombinant vectors. The result of recombinant vector was identified using PCR and DNA sequencing. Results: HGF cDNA ex- pression vector was constructed successfully. Conclusion: Constructing HGF cDNA expression vector may provide an experimental basis for the further study of the relationship between HGF and hepatic carcinoma.
