桉叶油对大鼠胚胎发育的影响 被引量:4

摘要 目的探讨桉叶油对SD大鼠胚胎发育的影响。方法取SD孕鼠25只,随机分成3个实验组(桉叶油高、中、低剂量)、溶剂对照组(花生油)和阳性对照组(环磷酰胺),每组5只。实验组和溶剂对照组分别在大鼠妊娠第10天用300、200、100mg/kg的桉叶油和花生油2ml/只灌胃,每天1次,连续5d。阳性对照组于孕第13天腹腔注射环磷酰胺1次(12.5mg/kg)。各组孕鼠于孕19d处死取胚胎,记录孕鼠的体重、子宫重、卵巢重和胎盘重;统计胚胎植入的总数、吸收胎数、活胎数和死胎数;观察胚胎外形,并测量胎鼠体重、身长和尾长。结果桉叶油灌胃SD孕鼠后,孕鼠的体重、子宫重、卵巢重及胎盘重与溶剂对照组比较均无显著性差异(P>0.05)。胚胎植入总数、吸收胎数、活胎数及死胎数与溶剂对照组相比均无显著性差异(P>0.05)。桉叶油各组活胎鼠平均体重、身长及尾长均较溶剂组高,但只有高剂量桉叶油组胎鼠平均体重(2.318±0.606)与溶剂组(1.462±0.397)比较有明显差异(P<0.05),其余指标与溶剂组比较无明显差异(P>0.05)。阳性对照组胎鼠平均体重、尾长明显低于溶剂组,且有差异(P<0.05)。阳性对照组胎鼠平均身长低于溶剂组,但统计学分析无差异(P>0.05)。结论本实验条件下,桉叶油对SD孕鼠胚胎无毒性,环磷酰胺对大鼠胚胎有毒性。 Objective To study the effects of eucalyptus on embryonic development of rats. Methods Totally 25 pregnant SD rats were randomly divided into 5 groups, each had 5 rats: three experiment groups ( high,medium,low doses of eucalyptus) , negative control group ( peanut oil) , positive control group (cyclophosphamide). Three experiment groups and negative control group were lavaged from gestation 10 to 14 days with eucalyptus in 100mg/( kg·d), 200mg/( kg·d), 300mg/( kg·d) and peanut oil 2ml per animal per day respectively, while positive control group was ip with 12.5 mg/( kg·d) cyclophosphamide at gestation 13 days. All pregnant rats were sacrificed at the 19th day. The body, uterus, ovary and placenta weights of the pregnant rats were measured respectively. The numbers of absorbed, live and dead fetus were counted, respectively. The length for fetus body and tail as well as body weights were measured. Results After administration of eucalyptus (100mg/kg,200mg/kg,300mg/kg) for 5 days, the body, uterus, ovary and placenta weights of pregnant rats showed no significant difference compared with negative control group (P 〉 0.05 ). The numbers for absorbed, live and dead fetus also showed no significant difference compared with negative control group ( P 〉 0.05 ). All of the fetus body weights, body length and tail length were larger in the treated groups than those in the negative control group. But only in high doses of eucalyptus groups, the fetus weight of body showed difference compared with negative control group (P 〈 0.05) , others had no significant difference (P 〉 0.05 ). The fetus body weight, tail length in the positive control group were lower than those in the negative control group (P 〈 0. 05) ,but the fetus body length showed no significant difference compared with negative control group (P 〉 0. 05). Conclusion Under the experimental condition, eucalyptus showed no embryonic developmental toxicity, where cyclophosphamide showed embryonic developmental toxicity.
