摘要
目的动态研究现场剂量碘化钾(KI)、碘酸钾(KIO3)对小鼠血细胞DNA损伤情况,进一步探索食盐加碘在剂型、剂量方面的安全性。方法将160只昆明种小鼠,按体重随机分为4组:(1)KI适碘剂量组(I-50μg/L,正常对照组);(2)KIO3适碘剂量组(I-50μg/L);(3)KI现场剂量组(I-180μg/L);(4)KIO3现场剂量组(I-180μg/L)。喂养20、30周时,每组取材20只,用单细胞凝胶电泳方法观察小鼠血细胞DNA损伤情况。结果 30周与20周相比各组别小鼠血细胞的拖尾均增长;20周时,与KI适碘剂量组相比,各实验组Tail Extent Moment指标差异显著(P<0.01);30周时,与KI适碘剂量组相比,各实验组Tail Extent Moment指标差异显著(P<0.01)。结论在KI、KIO3现场剂量的长期(30周)作用下,可以造成小鼠血细胞DNA损伤。
Objective To study the DNA damage on the mice blood corpuscle by the scene(population) dosage of KI and KIO3 in order to explore the safety of iodized salt within different iodine forms and dosages.Methods One hundred and sixty Kunming mice were randomly divided into four groups by weight(iodine concentration expressed in actual iodine content):(1) KI adequate group(I^-50 μg/L);(2) KIO3 adequate group(I^-50 μg/L);(3) KI scene-dose group(I^-180 μg/L);(4)KIO3 scene-dose group(I^-180 μg/L).After the mice were fed for 20,30 weeks(w),blood of 20 mice each group was taken for Single Cell Gel Electrophoresis(SCGE) determination and the DNA damage observation on blood corpuscle.Results From the Fluorescence Intensity Profile,the tail length of 30 w groups were all longer than that of 20 w groups.Whenever in 20 or 30 week,compared with KI adequate group,Tail Extent Moment in other three groups had remarkable change(P〈0.01).Conclusions Under long-term(30 w) effects of the KI and KIO3,scene-dose of them can cause DNA damage on blood cells in mice.
