摘要
目的研究灯盏花素对博莱霉素A5致肺纤维化大鼠肺组织中转化生长因子-β1(TGF-β1)及细胞外基质透明质酸(HA)含量的影响。方法将72只健康SD大鼠随机分为正常组、模型组、灯盏花素对照组和灯盏花素治疗组。除正常组及灯盏花素对照组外,其余2组大鼠经气管注入博莱霉素A5复制肺纤维化大鼠模型,各组于造模后d1分组予以给药,于给药7、14、28d分批处死各组大鼠,每次每组6只。应用ELISA法检测肺组织中TGF-β1及HA的含量。结果与模型组比较,灯盏花素治疗组病理改变与模型组表现一致,但较模型组减轻;在不同时段灯盏花素治疗组TGF-β1及HA含量均较模型组明显降低,但仍高于对照组和灯盏花素对照组。结论灯盏花素可能通过降低TGF-β1和HA含量,减轻博莱霉素A5致大鼠肺纤维化作用。
Aim To investigate the effect of breviscapine on TGF-β1 and hyaluronic acid of lung tissue in pulmonary fibrosis rats.Methods Seventy-two rats were divided into four groups randomly,including control group,model group,breviscapine control group and breviscapine treated group.On experimental day 0,the rats were intratracheally instilled with bleomycin,and then treated with breviscapine 10 mg·kg^-1·d^-1 until they were killed.On days 7,14 and 28 after instillation,six rats of each group were killed and the lungs were harvested for examination.TGF-β1 and hyaluronic acid of lung tissue were detected by ELISA method.Results The pathological lesion was lightened in breviscapine treated group compared with BLM group.The content of TGF-β1 and hyaluronic acid in lung tissue decreased significantly at different intervals of time in breviscapine treated groups and still to remain high in normal group and breviscapine control group.Conclusion Breviscapine may reduce the content of TGF-β1 and hyaluronic acid of lung tissue and may alleviate the process of rat pulmonary fibrosis induced by bleomycin.
