庆大霉素对兔成骨细胞增殖及超微结构的影响 被引量:12

Effects of Gentamycin on Proliferation and Ultrastructure of Primary CulturedRabbit Osteoblasts

摘要 目的观察庆大霉素对兔成骨细胞增殖及超微结构的影响。方法从出生2周的新西兰兔颅盖骨获取成骨细胞,在含不同浓度庆大霉素的DMEM培养液中培养24小时或72小时,采用四唑盐比色法观察庆大霉素对成骨细胞增殖的影响,并对在含不同浓度庆大霉素的DMEM培养液中培养96小时的成骨细胞进行透射电镜观察。结果高浓度庆大霉素组(800g/ml、1600g/ml)光吸收值明显低于对照组(P<0.01),高浓度庆大霉素组超微结构表现为内质网扩张,胞浆内见大量的吞噬溶酶体。结论高浓度庆大霉素明显抑制成骨细胞增殖,呈现一定的毒性作用。 Objective To observe the effects of gentamycin on the proliferation and ultrastructure ofprimary cultured rabbit osteoblasts. Methods Osteoblasts isolated from the two-week newly born rabbits werecultured for 24 h or 72 h in DMEM media with different concentrations of gentamycin- MTT method was used toobserve the effect of gentamycin on cell proliferation. The ultrastructural changes of cultured osteoblasts exposedto gentamycin for 96 h were also observed under transmission electron microscope. Results The opticaldensity values of higher concentration gentamycin groups (800 g/ml, 1600 g/ml) were much smaller thanthat of the control group ( P < 0. 01 ) . The swelling of mitochondria and the presence of abundant phagolyso-somes were characterized as the ultrastructural features. Conclusion High concentration gentamycindemonstrated a certain toxic inhibitive effects to osteoblasts.[
