摘要
目的:探讨L-肌肽预处理对急性心肌缺血大鼠心肌细胞凋亡相关蛋白表达的影响。方法:30只SD雄性大鼠随机分为L-肌肽预处理组、心肌缺血2h组及假手术组,采用冠状动脉结扎的方法建立大鼠心肌缺血模型。用TUNEL法观察各组大鼠的心肌细胞凋亡情况;采用Western blot检测各组大鼠心肌细胞bcl-2及caspase-3蛋白的表达水平,并探讨它们之间的关系。结果:与心肌缺血2h组相比,L-肌肽预处理组bcl-2蛋白的表达水平明显升高,心肌细胞凋亡数与bcl-2蛋白表达量值之间呈明显的负相关(r=-0.93,P<0.05);L-肌肽预处理组caspase-3蛋白的表达水平降低,心肌细胞凋亡数与caspase-3蛋白表达量值之间呈明显的正相关(r=0.89,P<0.05)。结论:L-肌肽对心肌缺血大鼠模型心肌梗死有明显保护作用,其作用机制可能与调节心肌细胞凋亡相关蛋白作用有关。
Objective To investigate the effects of L-carnosine preconditioning on the expressions of apoptosis-related proteins in myocardial cells of rats with acute myocardial ischemia (AMI). Methods Rat models of AMI were constructed by coronary artery ligation,and 30 SD male rats were randomly divided into AMI group,sham-operation group and L-carnosine-treated group. Terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) assay was used to analyze myocardial apoptosis and calculate the corresponding apoptosis indexes in all rats. And the protein levels of Bcl-2 and Caspase-3 in myocardial cells were analyzed by Western blot. Results As compared with AMI group,protein levels of bcl-2 and caspase-3 in L-carnosine-treated group were increased dramatically,and they were correlated with the myocardial apoptosis index positively (r=-0.93,P〈0.05) or negatively(r=0.89,P〈0.05),respectively. Conclusions L-carnosine has obviously protective effects against myocardial infarction in rats with AMI,which may be ascribed to its regulation on the expression of apoptosis-related proteins.
