摘要
目的初步研究苦苣菜水提物对糖尿病小鼠心肌组织超微结构变化的影响。方法用四氧嘧啶(A lloxan,ALX)构建糖尿病模型,分为6组,连续灌胃给药28d,1次/d。分别在第7、14、21、28d测小鼠空腹血糖值和体重,并于第28d取血测血清胰岛素,取心肌组织以透射电镜观察小鼠心肌组织超微结构的变化。结果干预结束后与模型组比较,苦苣菜水提物组小鼠心肌细胞肥大及坏死有所改善,细胞间质减少,肌丝排列相对整齐,苦苣菜水提物中剂量组改善较明显。结论苦苣菜水提物可以改善ALX糖尿病小鼠心肌超微结构,对小鼠糖尿病性心肌损伤具有保护作用。
Objective To investigate the effect of the water extract from Sonchus L(SOL) on the ultrastructural change of myocardial tissue in Alloxan-induced diabetic mice.Methods Diabetic models were induced by the i.v.injection of Alloxan(140mg/kg) in ICR mice and mice were divided into different groups,which were given SOL(4g/kg,2g/kg,1g/kg),Metformin or normal saline intragastrically once a day for four weeks.Blood glucose and body weight were monitored on the 7th,14th,21st and 28th day.The blood was taken for serum insulin checking.Myocardium was collected for observing the effects of SOL on the ultrastructural change of myocardial tissue.Results After the intervention,comparing with model group,the myocardial hypertrophy and necrosis were improved in SOL groups with decreased interstitial and neatly arranged muscle filament,especially in SOL medium group.Conclusion Water extract from Sonchus L can improve the injury of myocardium ultrastructure in Alloxan-induced diabetic mice,thus delayed occurrence and development of diabetic cardial injury.
