摘要
目的探讨凋亡相关基因HER2、P53在乳腺癌中的表达,及其与多药耐药基因P-gp、GST-π的关系。方法用Enivision免疫组织化学方法对66例未经化疗的乳腺癌手术切除标本进行HER2、P53及P-gp、GST-π的检测。结果乳腺癌组织中P53及P-gp、GST-π的阳性表达率各为50.0%、40.9%及42.4%,HER2的过度表达率为27.3%;乳腺癌中P-gp的表达与HER2的过度表达及P53、GST-π的阳性表达均呈正相关;HER2的过度表达与P53的表达具有相关性。结论凋亡相关基因HER2、P53过表达可能通过凋亡抑制作用,共同参与了乳腺癌的多药耐药。
Objective To detect the expression of apoptosis correlated gene HER-2,P53 in breast cancer and their correlations to P-gp and GST-π.Methods Immunohistochemistry analysis was used to detect the expression of HER-2,P53,P-gp and GST-π in 66 cases of breast cancer without chemotherapy.Results The positive rate was respectively 50.0%,40.9% and 42.4% in P53,P-gp and GST-π protein in breast cancer tissue,and the overexpression of HER2 protein in breast cancer tissue was 27.3%.The expression of P-gp protein was correlated with the overexpression of P53,HER2 and GST-π protein.The expression of P53 protein was also correlated with the overexpression of HER2 protein.Conclusion Multidrug resistance of breast cancer may be mainly mediated by GST-π and P-gp.The overexpression of apoptosis correlated genes HER-2 and P53 may be involved in the mechanism of breast cancer multidrug resistance by inhibit of apoptosis.
