摘要
实验以茜素红(ARS)为荷移试剂,采用分光光度法研究了ARS与头孢哌酮(CPZ)形成络合物的光谱性质及其反应的适宜条件。实验结果表明:头孢哌酮在2.0~200μg/mL范围内遵循比尔定律,回归方程为:A=0.0036ρ+0.000095(:ρμg/mL),检出限0.75μg/mL,表观摩尔吸光系数2.64×104L·mol-1·cm-1,相关系数为r=0.9965,并初步探讨了反应机理。用实验方法对尿样及生物样品进行加标回收测定,回收率在95.4%~104.8%。
The visible spectral characteristics and the reaction conditions of complex of cefoperazone(CPZ) and alizarin red S(ARS) were investigated by spectrophotometry with ARS as a chromogenic reagent.The results showed that Beer's law was obeyed in the concentration range of 2.0~200 μg/mL for CPZ and the regression equation was A=0.0036ρ+0.000095(ρ:μg/mL) with the correlation coefficient R of 0.9965,the apparent molar absorptivity was 2.64×104 L·mol-1·cm-1 and the detection limit was 0.75 μg/mL.Furthermore,the reaction mechanism was also explored elementarily.The proposed method has been successfully applied to the determination of CPZ in urine and biological samples,and the results were satisfying.
