Flow cytometry(FCM) and immunofluorescence staining technique were used to quantitatively measure the DNA content and P 53 gene product expression from 18 cases of pleomorphic adenoma and 6 cases of carcinoma arising from pleomorphic adenomas,in order to detect the significance of quantitative analysis of two parameters applied for pathological diagnosis of pleomorphic adenoma and carcinoma arising from pleomorphic adenoma.The results showed that the pleomorphic adenoma possessed the properties of DNA diploid of normal lacrimal gland cells,but the carcinoma arising from pleomorphic adenomas were characterized by DNA aneuploidy.The significant difference of P 53 gene product expression between the two kinds of tumors was observed.DNA content was positively correlated to the quantity of P 53 gene product expression.It was demonstrated that the quantitative determination of DNA content and P 53 gene product expression may provide objective indexes for the pathological diagnosis of this tumors.
