肾透明细胞癌易感性与表皮生长因子基因-61G〉A多态性的相关性 被引量:3

The association between the -61G 〉 A polymorphism in the epidermal growth factor gene and susceptibility to renal clear cell carcinoma

摘要 目的探讨中国南方汉族人群中表皮生长因子(EGF)基因-61G〉A多态性与肾透明细胞癌(CCRCC)易感性的相关性。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法检测中国南方汉族295例患者和295例正常人群的EGF.61G〉A基因型,分层分析探讨罹患CCRCC的相关因素。结果与GG型携带者比较,GA型(OR=1.49,95%CI:1.05~2.12)或AA型(1.99,1.12—3.52)携带者发生CCRCC的危险性明显升高。分层分析显示当携带GA或AA型的患者年龄、性别、吸烟以及体质量指数大于23是发生CCRCC的危险因素(年龄:2.48,1.54~4.01;男性:1.56,1.03~2.36;吸烟:2.17,1.23—3.82;体质量指数:1.96,1.28~3.01)。结论EGF-61G〉A多态性可能与我国南方地区汉族人群CCRCC易感性有关。 Objective To investigate the association between the -61G 〉 A polymorphism in the epidermal growth factor (EGF) gene and susceptibility to renal clear cell carcinoma (RCC) in a southern Chinese population. Methods We obtained information on demographic factors by a personal interview. The polymorphism was analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) using genomic DNA isolated from 295 patients with RCC and 295 age- and sex-matched controis. Stratification analysis by age, sex, smoking, drinking and BMI status was performed. Results As compared with the -61GG genotype, individuals carrying GA genotype ( OR = 1.49, 95% CI = 1.05 - 2. 12) or AA genotype ( 1.99, 1.12-3.52) had an increased risk of RCC. Stratified analysis revealed that the increased risk was more pronounced among young people (2. 48, 1.54-4. 01 ), males ( 1.56, 1.03- 2. 36), smokers (2. 17, 1.23-3.82) and those with body mass index larger than 23 (1.96, 1.28-3.01). Conclusion EGF-61G 〉 A polymorphism may contribute to the etiology of RCC in a southern Chinese population.
