碘对小鼠骨髓树突状细胞前体表型及功能影响

Effects of iodine on phenotype and function of murine dendritic cells in mice

摘要 目的探讨碘对小鼠树突状细胞(dendritic cell,DC)前体表型及功能的影响。方法选雌性C57BL/6 J小鼠40只,按碘摄入量随机分为适碘组(NI)、低碘组(LI)、10倍碘过量组(10H I)、50倍碘过量组(50H I)4组,喂养8个月后检测小鼠DC表面分子表达和培养上清中白细胞介素-12(interleukin-12,IL-12)水平以及DC刺激T细胞的增殖能力。结果 LI组小鼠DC表面分子CD40+CD11 c+、CD80+CD11 c+、CD86+CD11 c+、MHC-II+CD11 c+在CD11 c+细胞中所占比例均低于NI组小鼠,差异均有统计学意义(P<0.05);50H I组小鼠DC表面分子CD40+CD11 c+、CD80+CD11 c+、CD86+CD11 c+、MHC-II+CD11 c+在CD11 c+细胞中所占比例均高于NI组小鼠,差异均有统计学意义(P<0.05);LI组小鼠DC培养上清中细胞因子IL-12浓度为(275.8±32.6)pg/mL,低于NI组小鼠的(548.7±51.3)pg/mL(P<0.01);50H I组小鼠DC培养上清中细胞因子IL-12浓度为(963.2±83.4)pg/mL,高于NI组小鼠(P<0.01);LI组的刺激指数(SI)为(5.23±1.96),低于NI组小鼠的(8.61±2.15)(P<0.05);50H I组小鼠的SI为(12.38±2.61),高于NI组小鼠(P<0.05)。结论摄入一定水平的碘可诱导小鼠DC前体的增殖和成熟,提高机体的免疫功能。 Objective To discuss the influence of dietary iodine intake on the phenotype and function of murine dendritic cells(DLs).Methods Forty female C57BL/6J mice were randomly divided into four groups according to the iodine intake:low iodine(LI),normal iodine(NI),10-folds excessive iodine(10 HI),and 50-folds excessive iodine(50HI).Eight months after the treatment,the mice were sacrificed and the mononuclear cells were isolated from bone marrow of the mice and cultured in RPMI medium 1640 containing different cytokines to generate DCs.The phenotypic molecules,the level of interleukin-12(IL-12) in the supernatant of cultures of DCs and the proliferation of allogenic T cells stimulated by DCs were analyzed.Results The percentages of CD40+,CD80+,CD86+ and major histocompatibility complex class II+ cells in CD11c+ cells of LI were significantly lower than those of NI.Except the percentage of CD40+ cells in CD11c+ cells,all the parameters were statistically increased in 50HI compared with the NI(P0.05 or P0.01).The level of IL-12 in the supernatant of culture of DCs in LI was considerably decreased(P0.01),while that of 50HI was significantly enhanced compared with the NI(P0.01).Stimulative index(SI) of LI was 5.23±1.96,which was statistically lower than that of the NI(8.61±2.15,P0.05).SI of 50HI(12.38±2.61) was much higher than that of the NI.There was no difference in SI between 10HI and NI.Conclusion The results demonstrate that high level of iodine intake could promote differentiation and maturation of murine dendritic cells and enhance the immune function in vivo.
