摘要
目的:探讨细胞间黏附分子-1(ICAM-1)K469E基因多态性与高血压患者颈动脉粥样硬化关系及血清中的ICAM-1(sICAM-1)对其影响。方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术以及酶联免疫吸附试验(ELISA)对201例高血压患者和106例对照者的细胞间黏附分子K469E基因多态性及血清中sICAM-1水平检测。结果:高血压组ICAM-1基因型及等位基因频率与对照组比较差异无统计学意义,但在高血压患者中KK基因型颈动脉内膜-中膜厚度(IMT)比E基因携带者高血压患者的厚。高血压患者血清中sICAM-1水平及IMT较对照组增加,并且血清中sICAM-1水平随着高血压患者IMT增厚而增加。结论:在高血压人群中,ICAM-1 K469E基因多态性与颈动脉粥样硬化有关,E等位基因可能是颈动脉硬化遗传易感性基因,而且sICAM-1与颈动脉IMT密切相关。
Objective: To investigate the relation between the K496E gene polymorphisms and essential hypertension(EH) with the carotid atherosclerosis,and study the correlation between the serum level of ICAM-1 and the carotid atherosclerosis.Methods: 201 hypertension patients and 106 healthy people were selected,PCR-RFLP was used to determine their K496E gene polymorphisms,the serum level of ICAM-1 was measured by ELISE.Results: There was no significant difference in the distribution of ICAM-1 genotypes and allelie freguencies between EH group and the health control group,but in the EH group the EH subjects with KK genotype of carotid artery intima-media thickness(IMT) had increased than E allele carriers.The sICAM-1 levels and IMT were significantly higher in hypertensive patients than those of in the control subjects,and the serum levels of ICAM had an increasing trend with the thickness of IMT.Conclusion: ICAM-1 K469E gene polymorphism is associated with carotid artery arteriosclerosis of hypertension,E allele may be a risk factor for carotid artery arteriosclerosis.The serum ICAM-1 levels in the essential hypertension patients obviously increases in the EH group.
