大孔树脂对黑莓花色苷的吸附与解吸特性 被引量:5

摘要 为得到纯度较高的黑莓花色苷,比较了6种大孔吸附树脂对黑莓花色苷的吸附纯化效果,研究了AB-8型大孔树脂对黑莓花色苷的静态吸附动力学曲线、动态吸附及洗脱曲线。试验结果表明:AB-8大孔树脂对黑莓花色苷具有较好的吸附与解吸能力,是纯化黑莓花色苷的最佳树脂类型;在25℃条件下,AB-8型树脂对黑莓花色苷的吸附平衡时间为4 h,解吸平衡时间为3 h;解吸时宜选用体积分数为75%的乙醇溶液。最佳的分离条件为:上样速率为1.6 ml/min,上样体积1 100 ml粗提液,3倍柱床体积的75%乙醇以1.0 ml/min洗脱速率洗脱,在此条件下产品的得率为1.22%,花色苷回收率75.45%,纯度86.50%。 To obtain highly purified anthocyanins from blackberry named Hull,The adsorption and desorption characteristics of 6 kinds of macroporous resins were carefully compared.The characteristics of static adsorption dynamics,dynamic adsorption and elution dynamics were investigated as well.The results showed that AB-8 macroporous resins exhibited the best performance.The equilibrium time of absorption and desorption was 4 h and 3 h at 25 ℃,respectively.75% ethanol solution was the optimum solution for the desorption of blackberry anthocyanins.The optimal purification conditions was as follows: the sample flow rate was 1.6 ml/min,the sample addition volume was 1 100 ml,75%ethanol of three bed volumes as elution phase,and the eluting rate was 1.0 ml/min.Under these conditions,sample yield was 1.22%,anthocyanins recovery yield was 75.45% and its purity was 86.50%.
