摘要
目的通过检测白细胞介素-2(IL-2)、肿瘤坏死因子-α(TNF-α)和诱导型一氧化氮合酶(iNOS)的含量,在免疫分子水平上探讨大鼠感染犬弓首线虫后免疫功能的变化,观察iNOS的表达情况。方法将80只大鼠随机分为两组,每组40只,实验组每只大鼠灌胃犬弓首线虫感染期虫卵约3000个(对照组大鼠仅等量生理盐水灌胃),分别于大鼠感染犬弓首线虫后第1、2、3、4、5周随机取实验组和对照组大鼠各8只,常规股动脉取血分离血清,用放射免疫法测定其IL-2、TNF-α的值,用分光光度法测定iNOS的OD值并计算iNOS活力。结果实验组IL-2含量在第1、2、3、4周高于对照组,差异有统计学意义(P<0.01);TNF-α含量在各期均高于对照组,差异有统计学意义(P<0.05);实验组iNOS的表达在各期均高于对照组,差异有统计学意义(P<0.01),iNOS含量随感染时间的延长而呈逐渐升高趋势。结论实验组IL-2、TNF-α含量均高于对照组,iNOS的表达与细胞因子密切相关,其表达呈逐渐升高趋势,推测与IL-2和TNF-α变化呈正相关,提示IL-2、TNF-α和iNOS可作为病情判断和治疗效果与预后的重要指标。
OBJECTIVE To investigate the changes in immune function of SD rats infected with Toxocara canis through testing the levels of interleukin-2(IL-2),tumor necrosis factor-α(TNF-α)and iNOS,and the expression of inducible nitricoxide synthase(iNOS) was observed.METHODS In this study,80 SD rats were randomly divided into two groups(40 SD rats per group).Each rat of observation group was infected with 3000 infective stage eggs of Toxocara canis,each rat in control group was administrated with isochoric isotonic Na chloride(2ml).Eight rats were randomly collected from the observation group and the control group respectively at the 1st,2nd,3rd,4th and 5th week.Blood was collected from femoral artery,then the serum was abstracted and the levels of IL-2,TNF-α were measured with radioimmunoassay and the level of iNOS was analyzed with spectrum.RESULTS The serum level of IL-2 in observation group was higher than that in control group at the 1st,2nd,3rd and 4th week(P〈0.01).The content of TNF-α in observation group was higher than that in control group in each stage(P〈0.05).The expression of iNOS in observation group was higher than that in control group in each stage(P〈0.01),and the content of iNOS in observation group gradually increased with prolonged infection time.CONCLUSION The serum levels of IL-2 and TNF-α in observation group are higher than that in control group in each stage.The expression of iNOS is closely correlated with the changes of cytokines and presents an upward tendency.It is positively correlated with the changes of IL-2 and TNF-α and suggests that IL-2,TNF-α and iNOS can be regarded as important indicators for the diagnosis,treatment and prognosis in patients with larva migrans.
