摘要
目的观察运动性疲劳时大鼠海马组织中γ-氨基丁酸(GABA)、乙酰胆碱(Ach)、5-羟色胺(5-HT)、去甲肾上腺素(NE)、多巴胺(DA)含量和DA/5-HT比值的变化及人参总皂甙对其的影响,研究运动性疲劳产生的中枢机制和人参总皂甙的调节作用。方法 45只雄性SD大鼠适应性训练后,随机分为正常组、模型组和人参总皂甙组,采用跑台训练法,建立大鼠慢性运动性疲劳模型,强化训练期间,人参总皂甙组灌胃人参总皂甙溶液0.07 g.kg-1.d-1,正常组、模型组灌胃等体积的生理盐水,高效液相色谱法测定各组大鼠海马中GABA、Ach、NE、DA、5-HT含量。结果模型组大鼠海马中GABA、5-HT升高,Ach、NE、DA含量及DA/5-HT比值明显下降,与正常组比较均有显著性差异(P<0.05);人参总皂甙组大鼠海马中Ach、NE、DA含量及DA/5-HT比值升高,而GABA、5-HT降低,与模型组比较均有显著性差异(P<0.05)。结论海马中GABA、5-HT升高,Ach、NE、DA含量及DA/5-HT比值下降,可能是运动性疲劳产生的中枢机制之一;降低GABA、5-HT含量,升高海马中Ach、NE、DA含量及DA/5-HT比值,可能是人参总皂甙抗疲劳的作用机制之一。
Objective To observe the changes on the contents of GABA, Ach,5 - HT, NE, DA and the ratios of DA/5 - HT in hippocampal tissue of rats with exercise fatigue, and to detect the impact of Total Saponins of Panax Ginseng (TSPG) on them, to study the centre mechanisms of exercise fatigue and regulate effects of TSPG. Methods 45 male SD rats were randomly divided into control group, model group and TSPG group. In the wake of adaptive exercise, the chronic exercise fatigue models were established through the way of treadmill. During the periods of exercise reinforcement, intragastrie administration were exercised in TSPG 0.07 g · kg^-1 · d^ - 1 in TSPG group. Meanwhile, the same volume physiologic saline were given in control groups and model groups, and the contents of GABA,Aeh,5 -HT, NE and DA in hippocampal tissue of rats were detected by high efficiency liquid chromatography. Results The contents of GABA,5 - HT increased ,whereas the contents of Ach, NE, DA and the ratios of DA/5 - HT decreased in models group ; Compared with the control group, they all have the significant difference ( P 〈 0.05 ) ; however the contents of GABA .5 - HT decreased but the contents of Ach, NE, DA and the ratios of DA/5 - HT increased in TSPG group; Compared with the models group they all have the significant difference ( P 〈 0.05). Conclusions The increase of contents of GABA,5 - HT and the decrease of the contents of Ach, NE, DA and the ratios of DA/5 - HT may be one of the centre mechanisms of exercise fatigue, the decrease of contents of GABA, 5 - HT , and the increase in the contents of NE, DA or the ratios of DA/5 - HT in hippocampal tissue may be one of the mechanisms of antifatigue by TSPG.
