摘要
利用高效薄层色谱法测定棉籽油中游离棉酚的含量。自动点样仪、自动展开仪、扫描仪均采用瑞士卡玛薄层色谱系统;使用含0.5%CMC-Na的硅胶G高效薄层板;以甲苯-甲酸乙酯-甲酸-丁酮(体积比50:8:1.5:1)为展开剂;饱和三氯化锑氯仿溶液为显色剂;扫描方式:双波长紫外反射法;检测波长:550nm,参比波长:750nm。结果表明棉酚对照品溶液点样量在0.080—1.28μg范围内与峰面积呈良好的线性关系,r为0.9949;平均回收率(n=6)为91.5%—100.2%。本法操作简单、灵敏,结果准确可靠。
The content of free gossypol in cottonseed oil was determined by HPTLC.The CAMAG TLC system consists of automatic spotting,automatic developing,automatic scanner,etc.The experimental conditions are as follows:silica gel thin layer with 0.5% CMC-Na,methylbenzene-ethyl formate-formic acid-butanone(50:8:1.5:1) as developing solvent,chloroform solution of saturated antimony trichloride as coloration.Quantitative measurement was made by dual wavelength reflect linear scanning of gossypol spot at wavelengths of λs 550nm and λR 750nm.The calibration curve was linear in the range of 0.080—1.28μg for free gossypol(r=0.9949).The average recovery(n=5)were 91.5%—100.2%.The method is simple,accurate and reliable.
