摘要 目的研究磷脂酰肌醇-3激酶/蛋白激酶B(PI3K/Akt)特异性抑制剂LY294002与姜黄素联合对体外培养的人膀胱癌EJ细胞的抑制作用。方法用MTT法,检测姜黄素单独或联合PI3K/Akt特异性抑制剂LY294002对人膀胱癌EJ细胞的抑制率;用流式细胞技术及Western blotting法,检测药物单独或联合作用对EJ细胞凋亡的影响。结果联合LY294002能够显著提高姜黄素对EJ细胞的抑制率;联合LY294002能够增加EJ细胞的凋亡水平。结论 LY294002通过促进细胞凋亡有效提高姜黄素对EJ细胞抑制作用的敏感性,通过抑制PI3K/Akt信号转导通路的激活,可明显提高姜黄素对膀胱癌的治疗效果。 Objective To study the anti - proliferation and apoptosis induction effect of curcumin combined with LY294002 on human bladder cancer EJ cells. Methods The growth inhibition of curcumin and LY294002 alone or in combination on bladder cancer EJ cells by MTT assay. Effects of curcumin and LY294002 alone or in combination with phosphatidylinositol - 3 - kinases ( PI3 K) inhibitor LY294002 on apoptosis of EJ cells were detected by flow cytometry (FCM). Curcumin and LY294002 alone or in combination regulated caspase -3 and PARP protein expression in EJ cells by Western blotting analysis. Results The inhibition ratio of proliferation of EJ cells was increased significantly by the combination application of curcumin with LY294002 ( P 〈 0. 05 ). The apoptotic ratio of EJ cells was increased by curcumin in combination with LY294002. Conclusion The tumor - killing effect of curcumin on bladder cancer EJ cells could be increased significantly by the combination application of inhibiting phosphatidylinositol - 3 - kinases/protein kinases (PI3K/Akt) signal transduction and improving apoptosis may be involved in its inhibitory effects.
