摘要
目的:探讨蛋白酶体活性抑制对大鼠海马tau蛋白的泛素化影响。方法:利用双侧海马注射蛋白酶体的特异性抑制剂——乳胞素分别进行蛋白酶体活性检测、免疫印迹、免疫共沉淀,以检测大鼠海马内tau蛋白的泛素化改变情况。结果:(1)乳胞素引起蛋白酶体活性在24和48h较对照组明显下降;(2)免疫共沉淀结果显示,蛋白酶体活性抑制使tau蛋白发生了异常泛素化。结论:乳胞素抑制了蛋白酶体活性并导致了大鼠海马tau蛋白的异常泛素化。
Objective To investigate the effect of proteasome inhibition on the ubiquitination of tau protein. Methods We injected bilaterally lactaeystin, a specific inhibitor of proteasome, into the hippocampus of the rats and employed proteasome activity assay, Western blotting and eo-immunoprecipitation to observe the ubiquitination changes of tau protein in rats' hippoeampus. Results We observed the following results in our experiment: (1) Laetacystin obviously inhibited the ehymotrypsin-like activity of proteasome at the 24th h and the 48th h; (2) Proteasome inhibition increased the ubiquitination of tau. Conclusion Proteasome inhibition leads to abnormal ubiquitination of tau protein in rats' hippoeampus.
