摘要
目的:建立测定松柏科植物中莽草酸含量的方法,确定莽草酸的最佳生产原料及最佳采集期。方法:采用高效液相色谱法测定7种松柏科植物针叶和嫩枝及3-10月采集的雪松松针中莽草酸的含量。色谱柱为Diamonsil C18(150mm×4.6mm,5μm),流动相为0.1%磷酸溶液-甲醇(90∶10),流速为1.0mL·min-1,检测波长为217nm,柱温为30℃。结果:莽草酸在4~40μg·mL-1浓度范围内与峰面积积分值呈良好线性关系(r=0.9996);平均回收率为97.9%,RSD=1.43%(n=9)。同期采集的松柏科植物针叶和嫩枝中莽草酸含量均以雪松为最高,分别达3.93%、1.66%;以5月份采集的雪松针叶中莽草酸的含量最高,达4.11%。结论:本方法操作简便,结果准确、可靠,可用于松柏科植物中莽草酸的含量测定。雪松松针可作为莽草酸的生产原料,4-6月为最佳采集期。
OBJECTIVE: To establish a method for the content determination of shikimic acid from coniferous plants, and to confirm the best raw material for shikimic acid and collecting season. METHODS: Shikimic acid from needles and branches of 7 kinds of coniferous plants was determinated by HPLC. Diamonsil C^( 150 mm^4.6 mm, 5 ^m) was used, the mobile phase consisted of 0.1% phosphorus solution-methanol (90: 10), the flow rate was 1.0 mL·min-1, the detection wavelengh was set at 217 nm and the column temperature was 30℃. RESULTS: The linear range of shikimic acid was 4-40μg.mL-l(r=0.999 6) with an average recovery of 97.9% (RSD=1.43%, n=9). The highest contents of shikimic acid in needles and branches from coniferous plants both collected from Cedrus deodara were 3.93% and 1.66% respectively. The content of shikimic acid from pine needles of C. deodara in May was the highest 4.11%. CONCLUSION: The method is convenient, accurate and reliable for content determination of shikimic acid. The pine needles of C. deodara can be used as raw material of shikimic acid and April to June is the best collecting season
