摘要 目的 初步研究弧菌福尔马林灭活条件,为弧菌灭活疫苗的生产工艺提供参数。方法对弧菌培养菌液添加不同浓度的福尔马林,于不同温度下测定灭活时间;采用摇瓶和发酵罐培养弧菌,在28℃比较灭活效果,并检测弧菌灭活疫苗原液的安全性。结果福尔马林对弧菌的最低抑菌浓度为0.025%,不同浓度的福尔马林及不同温度所需的灭活时间不同,福尔马林浓度越大,灭活时间越短,加热能缩短灭活时间。摇瓶和罐灭活效果检测显示,在28℃,0.3%的福尔马林24 h可完全灭活,弧菌二联灭活疫苗原液的安全性符合要求。结论 0.3%福尔马林在28℃灭活24 h,是比较理想的弧菌灭活方法。 Objective To preliminarily study the condition for inactivation of vibrio,and provide parameters for development of production procedure for inactivated vibrio vaccine.Methods The culture liquid of vibrio was added with formalin at various concentrations and determined for time needed for activation at various temperatures.The vibrio was cultured in shake-flask and fermenter at 28℃ respectively,based on which the inactivation effects were compared,and the safeties of bulks of inactivated vaccine were evaluated.Results The minimum inhibitory concentration of formalin to vibrio was 0.025%.The times needed for inactivation with formalin at various concentrations and temperatures were different,which were shortened with the increasing formalin concentration and the increasing temperature.The inactivation effects in shake-flask and fermenter showed that vibrio was completely inactivated with 0.3% formalin at 28℃ for 24 h,and the safety of inactivated bulk met the relevant requirements.Conclusion The inactivation with 0.3% formalin at 28℃ for 24 h was suitable for vibrio.
