摘要
目的:探讨衰老雌性大鼠性激素及其受体的变化情况和何首乌饮延缓卵巢组织衰老的作用机制。方法:D-半乳糖连续腹腔注射致亚急性衰老动物模型,造模后灌胃何首乌饮连续60天后,大鼠断头取垂体、卵巢。采用全自动微粒子化学发光免疫分析方法检测各组大鼠血清LH,E2,FSH的含量;原位杂交方法检测各组大鼠卵巢细胞FSHR和LHR的变化;West-ern Bloting法检测各组大鼠卵巢ER的变化。结果:同正常对照组相比,模型组大鼠垂体指数和卵巢指数均明显降低,血清E2含量明显降低,LH、FSH升高,ER、FSHR、LHR的表达明显降低,差异有统计学意义(P<0.05)。与模型组比较,何首乌饮组垂体指数和卵巢指数均明显增高,血清E2含量明显升高,LH、FSH降低,ER、FSHR、LHR的表达明显升高,呈现一定的剂量依赖性。结论:衰老时大鼠卵巢正常功能被破坏,何首乌饮通过对性激素和其受体的调节而延缓卵巢的衰老。
Objective:To explore the changes of sex hormones and their receptors and the mechanism of polygonum multiflorum drink in delaying aging of ovarian tissues in rats.Methods:The rat models of acute aging were established by continuous intraperitoneal injection of D-galactose,after intragastric administration of polygonum multiflorum drink for 60 days,the rats were executed by decollation,their pituitary and ovary were obtained;automatic microparticle chemiluminescence immunoassay was used to detect the contents of LH,estradiol and FSH in serum;the changes of FSHR and LHR in ovarian cells were detected by hybridization in situ;Western blotting was used to detect the changes of ER in ovary.Results:Compared with normal control group,pituitary index and ovary index decreased significantly in model group,the serum content of estradiol decreased significantly,the expression levels of LH and FSH increased,the expression levels of ER,FSHR and LHR decreased significantly,there was significant difference between the two groups(P0.05);compared with model group,pituitary index and ovary index increased significantly in polygonum multiflorum drink group,the serum content of estradiol increased significantly,the expression levels of LH and FSH decreased,the expression levels of ER,FSHR and LHR increased significantly,showing a dose-dependent manner.Conclusion:The normal function of ovary is disturbed in aging rats,polygonum multiflorum drink can delay aging of ovary by regulating sex hormones and their receptors.
