摘要
目的:研究mTOR特异性抑制剂RAD001对胃癌SGC7901细胞增殖的影响,并探讨survivin在其中的作用。方法:常规培养胃癌SGC7901细胞,RAD001单独或联合顺铂干预,MTT法测定RAD001对胃癌SGC7901细胞增殖的影响,流式细胞术检测细胞凋亡率,免疫细胞化学和Western-blot检测药物作用后细胞中survivin蛋白的表达。结果:RAD001单独或联合顺铂干预后,①SGC7901细胞增殖受到抑制,抑制效应呈剂量依赖性并随时间的延长逐渐升高;②高浓度RAD001可以促进SGC7901细胞的凋亡;③SGC7901细胞中survivin蛋白表达下降。结论:RAD001可以通过下调细胞中survivin蛋白的表达抑制胃癌SGC7901细胞增殖,促进其凋亡。
Objective: To investigate the role of mTOR specific inhibitor RAD001 in inhibiting gastric cancer cell SGC7901 prolif-eration and the possible mechanism of survivin in the inhibition.Methods:SGC7901 cells were cultivated,treated with RAD001 alone or in combination with cisplatin.MTT,flow cytometry,Immunocytochemistry and Western-blot array were used to investigate the cell proliferation,apoptosis and the expression of survivin in cells.Results:After treated with RAD001 alone or in combination with cisplatin,the cell proliferation were inhibited in a dose-dependment manner,and the inhibition rates increased with the treat-ment time extending.High concentration of RAD001 could induce apoptosis of SGC7901 cells.The expression of survivin in SGC7901 cells decreased after treatment.Conclusion:RAD001 could inhibit the cell proliferation of SGC7901 and induce the apoptosis through down-regulating the expression of survivin.
