补肾填精方对再障模型小鼠骨髓造血干(祖)细胞CFU-GM、CFU-E的影响 被引量:3

Effect of Formula of Invigorating the Kidney for Essence Replenishment on CFU-E and CFU-GM in Mice Model of Aplastic Anemia

摘要 目的:观察补肾填精方对再生障碍性贫血(简称再障)模型小鼠骨髓造血功能的影响。方法:小鼠在接受给予环磷酰胺及苯油1周后每天经灌胃分别给予补肾填精方悬液3个剂量(20 mg.kg-1.d-1、30 mg.kg-1.d-1、40 mg.kg-1.d-1)35天后眶动/静脉采血法检测外周血细胞数,取小鼠左侧股骨骨髓制备骨髓有核细胞悬液并进行有核细胞(BMNC)计数,甲基纤维素半固体法测定红系祖细胞集落(CFU-E)及粒单系祖细胞集落(CFU-GM)。结果:模型组小鼠红细胞(RBC)、血红蛋白(Hb)、白细胞(WBC)、血小板(PLT)、BMNC、CFU-E及CFU-GM数目较空白组明显减少(P<0.01),中药中、高剂量组上述指标较模型组升高(P<0.05)。结论:补肾填精方具有促进骨髓造血干/祖细胞造血,提升外周血细胞作用。 Objective: To observe the effect of Formula of Invigorating the Kidney for Essence Replenishment on bone marrow haematogenesis in mice models of aplastie anemia. Method : After 1 week of accepting eyclophosphamide and benzene oil, the mouse were given to different doses of Formula of Invigorating the Kidney for Essence Replenishment (20mg/kg · d, 30mg/kg · d and 40 mg/kg · d). After 35 days peripheral blood cells were detected by blood collection method in arcula artery/vein, karyocyte was counted by taking mouse left femoral bone to prepare marrow karyocyte cell suspension, and CFU - E and CFU - GM were detected by semisolid cellog- ran method. Result: RBC, Hb, WBC, PLT, BMNC, CFU - E and CFU - GM were decreased in model group than in blank control group markedly ( P 〈 0. 01 ). Above indicators in High and middle doses of treatment groups were increased than those in model group ( P 〈 0.05 ). Conclusion : Formula of Invigorating the Kidney for Es- sence Replenishment can promote haematogenesis in haemopoietic stem cell, and improve cytosis of peripheral blood cells.
