摘要
目的探讨红细胞溶血实验(Red blood cell test,RBC)检测农药眼刺激性的适用分级标准和作为兔眼刺激性实验(Draize test)替代方法的适用性。方法选取29种市售农药进行RBC实验,并同步进行Draize实验。根据2种分级标准,将RBC实验结果与Draize实验进行相关性和一致性分析。结果按化妆品原料分级标准,区分无/轻刺激性和中度及以上眼刺激性农药的结果为:M cNemar-Bowker W=9.35(P=0.22);Kappa=0.55(P=0.002);Spearman R=0.58(P=0.001);按化妆品产品分级标准,区分无/微刺激性和轻度及以上眼刺激性农药的结果为:M cNemar-Bowker W=10.40(P=0.02);Kappa=0.54(P=0.001);Spearman R=0.61(P<0.001)。结论 RBC实验可参照化妆品原料分级标准检测农药眼刺激性;尚不能完全替代Draize实验,但可作为筛检农药眼刺激性的替代方法纳入到农药眼刺激性检测策略中。
Objective To select a better classification standard of RBC hemolysis test for predicting the eye irritancy of pesticides and to use RBC hemolysis test to replace the method described in the in vitro toxicology INVITOX Protocol N°37.Methods 29 kinds of pesticides were tested by RBC hemolysis test and Draize test at the same time.The classifications of the two tests were analyzed on their relevance and consistency.Results The result of RBC hemolysis test based on the first classification standard of cosmetic ingredients were:McNemar-Bowker W=9.35(P=0.22);Kappa=0.55(P=0.002);Spearman R=0.58(P=0.001).While based on cosmetic ingredients:McNemar-Bowker W=10.40(P=0.02);Kappa=0.54(P=0.001);Spearman R=0.61(P〈0.001).Conclusion RBC hemolysis test could be chosen to test the eye irritancy of pesticides based on the cosmetic standard.The results of RBC hemolysis test and Draize eye irritation test were relevant and consistent.As a screening eye irritation for pesticides,RBC hemolysis test could be an effective alternative method to Draize eye irritation test.
