利用谷蛋白分析小麦强优势组合亲本遗传差异

Parental Diversity of the Crosses with Strong Heterosis in Wheat Based on Glutenin Electrophoretic Patterns

摘要 利用SDSPAGE方法对小麦(黑麦)异源重组系异源2号组配的22个强优势杂交组合的亲本进行了谷蛋白分析。结果表明,谷蛋白图谱能将所有亲本区分开。统计了电泳分离出的HMW亚基和部分LMW亚基谱带,共25条相对迁移率不同的谱带,其中,24条具多态性。父本异源2号与其强优势组合母本问谷蛋白遗传距离较大,平均值为0.54。聚类结果表明,所有亲本可划分为两大类,异源2号可单独划分为一亚类。谷蛋白遗传距离与亲本各性状表型差异及F1各性状杂种优势间相关均不显著,难以用于预测杂种优势。 The storage protein glutenins of the parents of 22 strong heterotic combinations inwhich Yiyuan 2, a wheat-rye recombinant was used as male parent, were analyzed by sodium dodeeyl sulphate- polyacrylamide gel electrophoric (SDS- PAGE ) method. The resuits show ed that all parents could be differentiated according to the glutenin binding patterns. The bands of all high-molecular-weight(HMW) subunits and some of low-molecular weight(LMW) subunitswere surveyed. A total of 25 bands with different relative mobility were found by electrophoresis. Of them, 24 bands had polymorphisms. The Glutenin genetic distance(GD) between the male parent Yiyuan 2 and the female parents of its strong heterotic crosses were relatively greater, with an average of 0. 54. According to cluster analysis based on glutenin GD, all parents could be clustered as two groups. Yiyuan 2 could be classified as one sub-group by itself. The results also showed that the glutcnin GD was not significantly correlated with the parental genotypic diversity and F1 heterosis of each character. It was considered that the glutenin GDwas difficult to be used to predict heterosis.
