摘要
[目的]研究高效液相色谱法(HPLC)测定动物血制品中三聚氰胺的最佳条件。[方法]用1%三氯乙酸溶液、乙腈超声提取样品;经SPE柱净化后,进行高效液相色谱分析。[结果]最佳色谱条件为色谱柱Zorbax SB-C8;流动相为离子对缓冲液/乙腈=95/5(V/V);流速1.0 ml/min;柱温25℃;紫外检测器检测波长235 nm。三聚氰胺浓度为1.0~50.0 mg/L,线性相关系数为0.999 4,最低检出浓度为0.1 mg/kg,平均回收率为97.60%~100.65%,相对标准偏差为1.23%~3.04%。[结论]该方法快速简便,结果准确,重现性好。
[Objective]To investigate the optimumdetermination condittons of melamine in creatural blood produce using high performance liquid chromatography(HPLC).[Method]After extracted with ultrasonic in 1% trichloroacetic acid and acetonitrile,and purified by SPE,the melamine in each sample was determined by HPLC system.[Result]The optimum conditions of HPLC were as follows:the column of chromatogram Zorbax SB-C8,mobile phase ion-pairs buffer/acetonitrile=95/5(V/V),flow rate 1.0 ml/min,column temperature 25 ℃,UV-235 nm detector.The linear range was from 1.0 to 50.0 mg/L as the linear correlation coefficient was 0.999 4,the minimum detection limit of melamine was determined to be 0.1 mg/kg,the average recoveries of the method were 97.60%-100.65%,and the RSD were 1.23%-3.04%.[Conclusion]This method is reliable,simple,quick and accurately.
