摘要
目的:研究大鼠冠状动脉结扎时心肌组织中凋亡相关基因的表达状况与急性心肌缺血的关系。方法:结扎大鼠左冠状动脉前降支建立急性心肌缺血模型。描记Ⅱ导心电图;检测血清肌酸激酶(creatine kinase,CK) 活性、心肌组织丙二醛(malondialdehyde, MDA)含量和超氧化物歧化酶(superoxide dismutase,SOD) 活性;RTPCR 法检测凋亡相关基因Fas、CPP32 、Bcl2 和Bax 在心肌组织的相对表达量。观察伪结扎组、结扎组、苦豆碱(aloperine,Alo)给药组(20 mg·kg- 1)、普萘洛尔(propranolol,Prop)给药组(2 mg·kg-1) 对上述各指标的影响。采用单因素方差分析(One Way ANOVA) 对结果进行统计处理。结果:结扎大鼠左冠状动脉前降支可导致心电图明显的缺血性改变及血清CK 活性显著升高;心肌组织的MDA 含量和SOD 活性显著增加;缺血心肌组织中Fas、CPP32 和Bcl2基因的表达显著增加( P<0 .001),Bax 变化不明显( P< 0.05) 。Alo 和Prop 能明显缓解心电图的缺血性改变并降低血清CK 活性;
Objective: To study the expression of some apoptosis related genes in AMI and the influences of aloperine and propranolol. The relationship between apoptosis related genes and AMI were also investigated. Methods: AMI rat model was induced by ligation of the anterior branch of the left coronary artery. All animals were divided into four groups: sham operation group, occlusion group, aloperine group (20 mg·kg -1 ) and propranolol group (2 mg·kg -1 ). ECG lead Ⅱ was recorded. The level of serum creatine kinase (CK) activity, myocardial malondialdehyde (MDA) content and myocardial superoxide dismutase (SOD) activity were also determined. The myocardial expression of apoptosis related genes: Fas, CPP32, Bcl 2 and Bax were detected with RT PCR. Statistical significance in multiple comparisons was determined by One Way ANOVA. Results: Coronary occlusion of the rats led to significent ischemic changes of ECG, and increased the level of serum CK activity, myocardial MDA content and SOD activity. The myocardial expression of Fas, CPP32 and Bcl 2 were increased by coronary occlusion. But had no significance was detected in Bax expression. Aloperine and propranolol attenuated the ischemic changes of ECG, reduced the level of serum CK activity, myocardial MDA content and SOD activity. Both drugs also reduced the myocardial expression of Fas, CPP32 and Bcl 2, but no significance with Bax expression. Conclusion: The acute myocardial ischemic injury induced by ligation of the anterior branch of the left coronary artery may have relationship with apoptosis related genes: Fas, CPP32 and Bcl 2 family.
