摘要
目的:观察烟草浸提液(smokeless tobacco extract,ST)对大鼠成骨细胞(rat osteoblasts,ROBs)在钛板上附着和增殖的影响。方法:采用酶解-组织块法进行ROBs的原代培养,观察细胞生长状态;用碱性磷酸酶(ALP)组织化学染色法、矿化结节茜素红染色法对成骨细胞进行鉴定。不同浓度的ST作用于附着在钛板上的ROBs,观测细胞的铺展面积,MTT法检测钛板表面细胞的附着、增殖情况。采用SPSS 13.0软件包对数据进行双因素方差分析。结果:ALP染色显示,ROBs细胞内有黑色颗粒,矿化结节茜素红染色为红色,证明该细胞为成骨细胞。随着ST浓度的升高,ROBs在钛板上的铺展面积逐渐变小;增殖呈下降趋势,ST实验组与阴性对照组之间存在显著差异(P<0.05)。各实验组内前3 d存在显著差异(P<0.05),但3.2~50g/L组第4~7天间的差异无统计学意义(P>0.05)。结论:ST可影响ROBs在钛板上附着和增殖,并随ST浓度的增加而增强,提示吸烟不利于口腔种植的骨整合。
Objective:To investigate the effects of smokeless tobacco extract(ST) on the attachment and proliferation of Rat's Osteoblasts(ROBs) on titanium.Methods:ROBs were obtained from explants of rat skull tissues using enzyme-tissue-explants culture technique and identified by histochemical staining of alkaline phosphatase(ALP) and alizarin red staining.ROBs were cultured on titanium in the presence of ST at various concentrations,a computer analysis system was used to detect and analyze the shapes of ROBs attached to titanium,the attachment and growth of cells on titanium were measured by MTT assay.Statistical analysis was performed using SPSS 13.0 software package for two-way ANOVA.Results:ROBs were identified by histochemical staining of ALP and alizarin red staining.With the increasing of ST concentration,the size of cells became smaller gradually.The proliferation of ROBs on titanium was inhibited by ST in a dose-and-time-dependerd maner in 3-day-culture(P0.05),but not in 4~7-day culture when ST concentrations were 3.2~50 g/L(P0.05).Conclusion:ST may inhibit the attachment and proliferation of ROBs on titanium.
