摘要
目的:建立反相高效液相色谱法,分别比较中药复方与单味五指毛桃根提取物中补骨脂素大鼠体内的药动学差别,探讨中药复方对药效成分体内过程的影响。方法:实验大鼠分别灌胃中药复方、五指毛桃根提取物后,测定不同时间点大鼠血浆中补骨脂素浓度,采用药动学程序3P97对数据进行统计学处理,进行药动学模型拟合并计算相关药代动力学参数。结果:复方及五指毛桃根中补骨脂素在大鼠体内均呈一室模型,复方中补骨脂素的吸收滞后时间(Lag time)长于单味药材,其吸收速率常数(Ka)小,吸收半衰期t1/2(ka)长,吸收速度较慢,使得复方两次达峰时间较晚;两者消除速率常数(Ke)、消除半衰期t1/2(ke)比较表明复方排泄较慢,在体内的作用时间稍长;曲线下面积复方大于五指毛桃根,吸收多于后者,生物利用度相对高,两者在Lag time、Tmax(1)及Tmax(2)上存在显著性差异。结论:中药复方与单味药材体内药动学之间存在差异,其产生的原因可能与复方间的相互配伍,复方成型工艺或其中添加辅料有关。
Objective: To compare the pharmacokinetics of psoralen in compound recipe and the extractive of hispid fig root,through Reversed Phase High Performance Liquid Chromatography(RP-HPLC) in rats respectively.To provide a new way to investigate the effect of combination in physiological disposition.Methods: The rats were intragastric administered with compound recipe and hispid fig root.The blood concentration of different time point were tested by the proposed established RP-HPLC and sample processing.The blood concentration data analyzed by the pharmacokinetics program 3P97 were used to determine the parameters of pharmacokinetics and the compart-mental model.Results: Psoralen in compound recipe and the extractive of hispid fig root showed one compartment model in rats.There were significant difference between compound recipe and hispid fig root in Lag time,Tmax(1) and Tmax(2).Compound recipe had slower absorption rate,higher AUC and bioavailability than hispid fig root.Conclusion: There is significant difference between compound recipe and hispid fig root on pharmacokinetics in vivo.The probably reason of them are intercompatibility,modeling technique and adjuvant of compound recipe.
