摘要
以吉林白鹅和籽鹅为试验对象,采用RT-PCR及免疫组织化学技术,检测28,56日龄鹅皮肤组织中IGF-Ⅰ基因表达和定位情况。结果表明:IGF-ⅠmRNA表达于上述两个日龄的皮肤组织中。IGF-Ⅰ特异性免疫染色明显表达于28日龄吉林白鹅和籽鹅胸部皮肤毛根鞘的扁平细胞和复层细胞,腹部皮肤内根鞘、外根鞘及毛小皮。在56日龄2鹅种胸部和腹部皮肤扁平细胞、复层细胞、皮质和髓质中也检测到IGF-Ⅰ蛋白免疫活性。在上述鹅种不同日龄不同组织细胞间IGF-Ⅰ蛋白免疫活性的表达强度明显不同。
In the experiment, with RT- PCR and immunohistochemistry, the expression and location of IGF mRNA in skin and hair follicle from different development phases of geese were detected. The results showed that the mRNA and protein of IGF- I were present in the above two tissues of two days. The specific immunostaining of IGF- I was mainly expressed in fiat cell and amb-layer cell of root sheath of chest skin tissues of 28 days Jilin white geese and Zi geese, and was detected in the internal root sheath, outer root-sheath and hair cuticle of skin of abdomen;the immunostaining of IGF- I was also detected in the flat cell and arab-layer cell cortex and medulla of chest and abdomen skin and hair follicle on 56 days, and the intensity of expression was distinct to the tissues and cells for the different days in the above geese.
