尼美舒利对人舌鳞癌裸鼠移植瘤环氧化酶-2的影响

摘要 目的研究环氧化酶-2(COX-2)抑制剂尼美舒利对人舌鳞癌裸鼠移植瘤的抑制作用,分析尼美舒利对COX-2基因及蛋白表达的影响,进一步探讨其抑癌机制。方法取人舌鳞癌Tca-8113细胞接种于16只裸鼠皮下,建立人舌鳞癌裸鼠移植瘤模型,随机分为对照组和尼美舒利治疗组,计算尼美舒利的抑瘤率。反转录-聚合酶链反应(RT-PCR)检测裸鼠移植瘤组织COX-2mRNA表达。结果尼美舒利治疗组抑瘤率为47.2%。尼美舒利治疗组裸鼠移植瘤组织COX-2mRNA水平均低于对照组,差异有统计学意义(P<0.05)。结论 COX-2抑制剂尼美舒利可下调COX-2基因表达,该作用可能是COX-2抑制剂抑制肿瘤血管生成从而抑制肿瘤生长的机制之一。 Objective To investigate the inhibitory effect of nimesulide(NIM),an inhibitor of cyclooxygenase-2(Cox-2),on human tongue cancer xenografts in nude mice and to analyze its impact on expressions of Cox-2 gene and protein,so as to further explore the mechanism that underlies its anti-cancer effect.Methods Human tongue cancer Tca8113 cells were isolated and implanted subcutaneously in 16 nude mice for establishment of human tongue cancer xenograft models.The subjects were then divided randomly into NIM therapy group and control group.The inhibitory rate of NIM was calculated.RT-PCR was used to detect the expression of Cox-2 mRNA in mice xenografts tissue.Results The inhibitory rate of tumor growth was 47.2% in NIM therapy group.The expression of Cox-2 mRNA in xenografts tumor tissue was significantly lower in NIM therapy group compared with control group,which reached the level of statistical difference(P0.05).Conclusion NIM could down-regulate the expression of Cox-2 gene,which may be one of the mechanisms that underlies the angiogenesis inhibition by Cox-2 inhibitor.
