摘要
目的探讨Caveolin-1与MMP-2在食管鳞癌组织中的表达及其临床意义。方法应用组织芯片技术结合免疫组织化学检测63例食管鳞癌组织,20例食管鳞状上皮非典型增生及15例正常食管鳞状上皮中Caveolin-1与MMP-2蛋白的表达。结果食管癌中Caveolin-1与MMP-2的表达高于正常食管黏膜、非典型增生食管黏膜(P<0.05);Caveolin-1的表达与淋巴结转移、病理学分级及临床分期有关,而与患者的年龄、性别,肿瘤大小、浸润深度无关(P>0.05);MMP-2的表达与淋巴结转移、浸润深度有关,而与患者的年龄、性别,肿瘤大小、病理分级及临床分期无关(P>0.05)。Spearman等级相关分析显示Caveolin-1与MMP-2表达呈正相关(r=0.390,P=0.002)。结论 Caveolin-1与MMP-2过表达与食管癌的发展、浸润及转移有关,Caveolin-1可能上调MMP-2的活性。
Objective To study the expression of Caveolin-1 and MMP-2 proteins in esophageal squamous cell carcinoma and its clinical significance.Methods Caveolin-1 and MMP-2 proteins were tested by immunohistochemistry and tissue microarray in 63 cases of esophageal squamous cell carcinoma,20 cases of atypical hyperplasia and 15 cases of normal esophageal squamous cell.Results The expressions of Caveolin-1 and MMP-2 were significantly higher in esophageal carcinoma than those in normal esophageal mucosa and esophageal dysplasia(P〈0.05);Caveolin-1 expression in esophageal carcinoma was correlated with lymph node metastasis,pathological grade and clinical stage,but not with age,gender,tumor size and invasive depth(P〉0.05).MMP-2 expression in esophageal carcinoma was correlated with lymph node metastasis and depth of invasion,but not with age,gender,tumor size,pathological grade and clinical stage(P〉0.05).Spearman rank correlation analysis showed that the expression of Caveolin-1 was positively correlated with MMP-2(r=0.390,P=0.002).Conclusion The over-expression of Caveolin-1 and MMP-2 is correlated with development,invasion and metastasis of esophageal carcinoma,and the Caveolin-1 might increase the activity of MMP-2.
