摘要
目的:探讨帕瑞昔布预处理对大鼠局灶性脑缺血再灌注损伤(Ischemia-reperfusion,I/R)的影响及其机制。方法:雄性SD大鼠64只,体重250~300 g,随机分为4组(n=16):假手术组(S组),单纯I/R组I,/R+帕瑞昔布5 mg/kg预处理组(P5组),缺血再灌注组+帕瑞昔布10 mg/kg预处理组(P10组)。线栓法制作大鼠大脑中动缺血2 h再灌注24 h模型,P5组、P10组在缺血前30 min分别经颈内静脉向向心端注入5 mg/kg和10 mg/kg帕瑞昔布注射液,S组及I/R组给予等容积生理盐水。在缺血2 h及再灌注24 h行神经功能缺失评分,氯化三苯四唑氮(2,3,5-Triphenyl tetrazolium chloride,TTC)染色观察脑梗死体积及体积百分比,HE染色观察海马CA1区神经元病理改变,放射免疫法检测脑组织中血栓素A2(Thromboxane,TXA2)、前列环素(ProstacyclinI2,PGI2)含量并计算TXA2/PGI2比值。结果:帕瑞昔布预处理能显著降低大鼠局灶性脑I/R后脑梗死体积及梗死体积百分比,改善神经功能,减轻缺血侧海马CA1区损伤,降低缺血侧额区皮质中TXA2含量及TXA2/PGI2比值。结论:帕瑞昔布预处理对大鼠局灶性脑I/R损伤具有保护作用,其机制可能与通过降低脑组织中TXA2/PGI2比值有关。
Objective:To investigate the effect and the mechanism of parecoxib pretreatment on brain against focal cerebral ischemia-reperfusion injury in rats.Methods:Sixty-four male SD rats weighing 250~300 g were randomly divided into 4 groups(n=16):groupⅠshame operation(S);groupⅡfocal cerebral I/R(I/R);group Ⅲ,Ⅳ parecoxib 5/10 mg/kg +focal cerebral I/R(P5,P10).Middle cerebral artery occlusion(MCAO)models were made by reforming Longa suture method in SD rats.At group Ⅲand groupⅣ,30 minutes before ischemia,inject 5 mg/kg and 10 mg/kg parecoxib injection through the internal jugular vein to the proximal end respectively.GroupⅠ and groupⅡreceived equal volume of normal saline.The infarct volume and the infarct volume fraction were detected by TTC staining,the pathological changes in CA1 region of hippocampus were detected by HE staining,radioimmunoassay technique was used to determine the content of TXA2/PGI2 in brain.Results:Pretreatment with parecoxib,the longa scores、the infarct volume and volume fraction、 the pathological changes in CA1 region of hippocampus、the content of TXA2 in brain and the ratio of TXA2/PGI2 were reduced.Conclusion:Pretreatment with parecoxib can protect the brain from focal cerebral I/R injury by reduced the content of TXA2 in brain and the ratio of TXA2/PGI2.
