摘要
目的探讨阿托伐他汀及氯沙坦预治疗对野百合碱诱发的大鼠肺动脉高压的作用。方法雄性Sprague-Dawley大鼠32只,体质量250~300g,随机均分为正常对照组、肺动脉高压组(PAH)、氯沙坦预治疗组(Los)和阿托伐他汀预治疗组(Ato)。后3组一次性腹腔注射野百合碱(MCT)60mg/kg,正常对照组予以等体积生理盐水腹腔注射。从野百合碱给药日起,Los组和Ato组大鼠分别给予氯沙坦50mg/(kg·d)、阿托伐他汀5mg/(kg·d)灌胃治疗,正常对照组及PAH组接受等体积生理盐水灌胃。4周后,测定右心室收缩压(RVSP)及右心室肥厚指数(RVHI)。运用图像分析仪分析肺组织切片,测定肺小动脉管壁厚度占管径的百分比(WT%)及管壁面积占血管总面积的百分比(WA%)。结果 Los组和Ato组的RVSP显著低于PAH组[Los组(30.5±4.6)mmHg,Ato组(30.1±3.4)mmHg比PAH组(44.7±2.4)mmHg,均P<0.01]。经氯沙坦或阿托伐他汀治疗后的大鼠,RVHI明显低于PAH组[Los组(36.4±4.3)%,Ato组(35.9±4.8)%比PAH组(42.8±5.0)%,均P<0.05]。与PAH相比,Los组与Ato组的WT%、WA%均明显降低[WT%:Los组(24.4±2.3)%,Ato组(22.5±2.4)%比PAH组(35.3±3.3)%;WA%:Los组(43.2±4.7)%,Ato组(41.0±4.4)%比PAH组(58.9±3.0)%,均P<0.01]。上述指标在Los组与Ato组间差异无统计学意义(P>0.05)。结论阿托伐他汀和氯沙坦能改善由野百合碱诱导的大鼠PAH及血管重构。
Objective To explore the effect of atorvastatin and losartan on monocrotaline-induced pulmonary arterial hypertension in rats. Methods Thirty-two Sprague-Dawley rats were randomly divided into four groups:normal control (Ctr group),untreated pulmonary arterial hypertension group(PAH),Losartan group [Los,treated with 50 mg/(kg·d) of losartan intragastrically after monocrotaline injection],and Atorvastatin group [Ato group,treated with 5 mg/(kg·d) of atorvastatin intragastrically after monocrotaline injection]. The pulmonary arterial hypertension was induced by intraperitoneal injection of 60 mg/kg monocrotaline. After 4 weeks,right ventricular systolic pressure (RVSP) and right ventricular hypertrophy index (RVHI) were measured. WT%(wall thickness to extennal diameter ratio) and WA%(wall to total area ratio) of pulmonary arterioles were evaluated by image analysis software. Results RVSPs in Los group and Ato group were significantly decreased than that in PAH group [Los(30.5±4.6)mm Hg,Ato(30.1±3.4)mm Hg vs PAH(44.7±2.4)mm Hg,P0.01 respectively]. In comparison with PAH group,RVHI in losartan group and atorvastatin group were also obviously reduced [Los (36.4±4.3)%,Ato (35.9±4.8)% vs PAH (42.8±5.0)%,P0.05 respectively]. WT% and WA% of pulmonary arterioles were significantly reduced by the administration of losartan or atorvastatin,as compared with PAH group [WT%:Los(24.4±2.3)%,Ato(22.5±2.4)% vs PAH(35.3±3.3)%;WA%:Los(43.2±4.7)%,Ato (41.0±4.4)% vs PAH(58.9±3.0)%,P0.01 respectively]. There was no significant difference between Los group and Ato group (P0.05). Conclusion Pulmonary arterial hypertension and arterioles remodeling induced.by monocrotaline in rats could be attenuated by the treatment with atorvastatin and losartan respectively.
