摘要 目的:建立测定人血浆中抗癫痫药拉莫三嗪的方法。方法:采用反相高效液相色谱法,以萘普生为内标,血浆样品以直接检蛋白测沉波淀长法为处28理0n。m色。谱结柱果为:拉X莫Te三rra嗪?R血P药18浓,流度动在相0.为5~甲3醇2.0-0m.0g1.mL-o1l.范L围-1磷内酸线二性关氢系钾良缓好冲(液r(=pH0.969.09)(9)6,6最:3低4)检,流测速浓为度为1.00m.2Lm.gm.iLn--11,,提取回收率为68.20%~82.35%,方法回收率为99.03%~103.21%,日内、日间RSD均<4.00%。结论:本方法操作简便,灵敏度高,重复性好,能快速准确测定人血浆拉莫三嗪浓度。 OBJECTIVE: To develop the method for the determination of lamotrigine in human plasma. METHODS: RP-HPLC was adopted. The samples were extracted by direct protein precipitation using naproxen as internal standard. XTerra RP18 column was used with the mobile phase consisted of methanol-0.01 mol .L^-1 potassium dihydrogen phosphate buffer(pH 6.0)(66:34). The flow rate was 1.0 mL. min-~, and the detection wavelength was set at 280 nm. RESULTS: The linear range of lamotrigine was 0.5~32.0 mg-L-t(r=0.999 9). The lowest detection concentration was 0.2 mg-L-~. The extraction recovery was 68.20%~82.35% and the method recovery was 99.03% ~103.21%. The RSD of intra-day and inter-day were both below 4.00%. CONCLUSION: The method is simple, sensitive and reproducible for concentration determination of lamotrigine in human plasma
