电针预处理抑制癫痫持续状态诱发的海马区神经元凋亡研究 被引量:6

Electroacupuncture Pretreatment Suppressed Hippocampal Neuron Apoptosis Induced by Experimental Epileptic Model in Rats

摘要 目的:观察电针预处理对癫痫持续状态(status epilepticus,SE)诱发的海马区神经元凋亡的影响。方法:20只SD大鼠被随意分为4组:对照组、癫痫组、电针组、电针对照组,电针组和电针对照组给予电针预处理3周后,按经典方法建立锂-匹罗卡品大鼠癫痫模型,用TUNEL法及免疫荧光法观察海马区神经元的凋亡。结果:(1)与对照组比较,癫痫组和电针对照组海马CA1/CA3区TUNEL及Caspase-3阳性的神经元明显增多(P<0.01,n=5);(2)与癫痫组和电针对照组相比,电针预处理减少了海马CA1/CA3区TUNEL及Caspase-3阳性神经元的数量(P<0.05,n=5)。结论:电针百会及大椎穴预处理,可以减低SE诱发的海马区神经元的凋亡。 Objective:To observe the suppressive effect of electroacupuncture(EA)pretreatment on hippocampal neuron apoptosis induced by Li-pilocarpine status epilepticus.Methods:20 SD rats were randomly divided into four groups:Control group,EA group,Pilo group and Placebo group.Pilo group and Placebo group rats were pretreated with electro-acupunture for 3 weeks.Li-pilocarpine status epileptic model was established according to the previous schedule.TUNEL and immunofluoresence were employed to observe the damaged and surviving neurons in hippocampus.Results:(1)Compared with control group,the Pilo and Placebo groups’ rats exhibited a large number of TUNEL-positive and caspase-3 positive cells in the CA1/CA3 region(P〈0.01,n=5).(2)EA pretreatment at Baihui and Dazhui acupoints decreased the total number of TUNEL-positive and caspase-3 positive cells in CA1/CA3 region of hippocampus in comparison with Pilo and Placebo groups(P〈0.05,n=5).Conclusion:EA pretreatment at Dazhui and Baihui acupoints suppressed the SE induced hippocampal neuron apoptosis in rats.
