DeBDE对学习记忆能力及BDNF基因表达的影响 被引量:1

Effects of Decabromodiphenyl Ether Exposure on Learning and Memory Ability and Hippocampal BDNF Gene Expression of Male Rat

摘要 目的研究十溴联苯醚(decabromod iphenylether,DeBDE)暴露对雄性大鼠学习记忆能力及海马组织中BDNF基因表达的影响。方法雄性SD初断乳大鼠经口灌胃给予不同剂量100,300,500 mg/(kg.d)的DeBDE染毒,对照组给予溶剂玉米油。60 d后用Morris水迷宫测试其学习记忆能力,并用定量PCR检测其大脑海马组织中脑源性神经生长因子的表达。结果高、中剂量组大鼠空间学习、记忆能力较对照组明显降低,差异有统计学意义;低剂量组大鼠学习记忆能力与对照组比较,差异无统计学意义;高、中、低剂量组大鼠海马组织中BDNF基因的表达都显著降低,且随染毒剂量增加表达有逐渐下降的趋势。结论 DeBDE可以损害大鼠的学习记忆能力,可能和降低海马BDNF的表达有关。 Objective To study the effects of decabromodiphenyl ether(DeBDE) on the learning and memory ability and on hippocampal BDNF gene expression of male rat.Methods Weaning male SD rats were given DeBDE at doses of 100,300 and 500 mg/(kg·d) by gavage for 60 days,and control rats were give solvent(corn oil) only for the same period.Then their learning and memory ability were tested by Morris water maze,and their hippocampal BDNF expression were evaluated by real-time PCR.Results Rats treated with 500 and 300 mg/(kg·d) DeBDE had significantly lower spatial acquisition and spatial memory than control rats;rats treated with 100 mg/(kg·d) DeBDE showed the same ability of spatial acquisition and memory as the control.All groups given DeBDE had lower expression of BDNF gene in hippocampal region of the brain,and there was a significant negative trend of BDNF gene expression over DeBDE doses.Conclusion DeBDE exposure compromises the learning and memory ability of male rats,which may be related to its effects on reducing hippocampal BDNF expression.
