摘要
目的研究蛋白酶体抑制剂MG132对体外培养的大鼠心肌细胞肥大的影响。方法采用[3H]-亮氨酸参入率测定心肌细胞蛋白质合成代谢,采用IPP 6.0图像分析软件分析心肌细胞表面积,以及用RT-PCR检测β-肌球蛋白重链(β-MHC)表达。结果 MG132预处理逆转了苯肾上腺素诱导的心肌细胞[3H]-亮氨酸参入率增加(P<0.01)、表面积增大(P<0.01)和β-MHC mRNA表达上调(P<0.05)。结论 MG132能够抑制心肌细胞肥大,具有心肌保护作用。
Objective To investigate the effects of proteasome inhibitor MG132 on cardiomyocyte hypertrophy in vitro.Methods Cardiomyocyte protein synthesize was determined by measuring the-Leucine incorporation rate.With the aid of IPP 6.0 Image software,the surface areas of cardiomyocytes was analyzed.The mRNA expression of fetal gene β-myosin heavy chains(β-MHC) was measured by reverse transcription-polymerase chain reaction(RT-PCR).Results MG132 significantly inhibited phenylephrine(PE) induced increase in cardiomyocyte-Leucine incorporation rate(P0.01),surface area(P0.01)and β-MHC mRNA expression(P0.05).Conclusion MG132 may inhibite cardiomyocyte hypertrophy,and demonstrate an cardioprotective activity.
