有机铵盐促进Gewald反应合成多取代2-氨基噻吩类化合物的研究 被引量:7

摘要 2-氨基-4,5-二甲基噻吩-3-乙酸乙酯类化合物是合成小麦全蚀病的高效防治药剂硅噻菌胺的重要关键中间体.为了高效合成这类关键中间体,发展了一种新型的合成方法,利用仲胺与对甲苯磺酸复合形成有机铵盐可以高选择性地促进Gewald反应合成相应2-氨基-4,5-二甲基噻吩-3-乙酸乙酯类杂环化合物,得到较为理想的分离产率和纯度,并深入考察了在该反应中各种不同因素的影响. 2-Amino-4,5-dimethyl thiophene-3-acetic ester is an important intermediate for Silthiofam,which is highly efficient pharmacy for wheat disease.This manuscript developed a novel synthetic process for the preparation of 2-amino-thiophene derivatives through aminium salt-promoted Gewald reaction.The desired products,substituted 2-amino-thiophene derivatives,have excellent chemoselectivity and high yields under mild conditions.
