摘要
目的探究海带多糖(Laminaria japonica polysaccharides,LJP)对慢性局部电离辐射致雄性大鼠睾丸细胞损伤的影响。方法将适应饲养7 d的56只Wistar雄性大鼠随机分为正常对照1组、辐射模型3组和LJP干预3组,每组8只;LJP干预3个组给予LJP的生理盐水溶液,其余各组以等量生理盐水灌胃;经灌胃处理7d后,除对照组外的其它6组,以60Coγ射线进行局部照射,除周末两天不照射外,每天1次,每周连续5次,共20次,总剂量每只2.3 Gy;分别在停止照射后1、7和14 d处死大鼠,分离睾丸;采用免疫组化、单细胞凝胶电泳、流式细胞术的方法研究海带多糖对电离辐射致大鼠睾丸组织caspase3蛋白表达量和睾丸细胞DNA损伤及其含量的影响。结果停止辐射1d、7d、14d后LJP各组与对应模型组相比,睾丸组织形态好于对应模型组,睾丸组织caspase3蛋白表达量降低,其彗星拖尾率、尾长减少;HC峰内细胞占睾丸细胞百分比明显低于对应模型组,且LJP 14d组1C、2C、4C峰内细胞占睾丸细胞百分比均显著高于模型14d组。结论 LJP可明显降低辐射所致大鼠睾丸组织caspase 3蛋白的异常表达和各睾丸细胞的DNA损伤,促进损伤细胞的修复。
Objective To investigate the effects of Laminaria japonica polysaccharides(LJP) on testicular cell in rats damaged by chronic local ionizing radiation.Method Fifty six rats were randomly divided into 7 groups: the normal control group,3 model groups and 3 LJP groups,8 in each.The rats in each LJP group were administered with 3 ml LJP solution dissolved in normal saline.Other groups were administered with equal quantity of saline.After 7 d intragastric administration,the model groups and LJP groups were irradiated locally with 60Coγ,once a day except the weekends for 20 times totally.Each rat was irradiated with the total dose of 2.3 Gy.The caspase 3 protein expression,DNA damage and the testis cells content in rats' testis were detected by immunohistochemistry,single cell gel electrophoresis and flow cytometry respectively.Results 1d,7d,14d after cessation of radiation,caspase 3 protein expression in testicular tissue was reduced in each LJP group compared with the corresponding model group,and the rate of comet tail and tail length also reduced.The percentages of cell within HC peak in LJP groups were significantly lower than those in the corresponding model groups.The percentages of cells within 1C,2C and 4C peak in 14 d LJP group were also increased significantly compared with the 14d model group.Its testicular morphology was better than that in the corresponding model group.Conclusion LJP may reduce the radiation-induced testicular cell DNA damage,inhibit the abnormal expression of caspase 3 protein and consequently slow down the testicular cells injury,and promote their recovery.
