摘要
目的应用蛋白质组学技术寻找丹参逆转人胃癌阿霉素耐药细胞SGC7901/ADR相关蛋白分子,探讨丹参逆转胃癌多药耐药机制。方法以人胃癌阿霉素耐药细胞SGC7901/ADR和丹参逆转后的SGC7901/ADR细胞为研究对象,用固相pH梯度等电聚焦二维聚丙烯酰胺电泳技术展示2种细胞表达的全蛋白,凝胶考马斯亮蓝染色,Image Master 2DMelanie 5.0图像分析软件分析并比较差异表达的蛋白分子。结果 SGC7901/ADR中检测到549个蛋白点,丹参逆转后SGC7901/ADR中检测到469个蛋白点,差异点87个,SGC7901/ADR细胞经丹参逆转后表达下调蛋白36个,上调蛋白51个。结论丹参逆转后SGC7901/ADR细胞中差异表达的蛋白质分子可能与丹参对胃癌多药耐药逆转机制相关。
Objective It is to search for the related proteins of radices salviae miltiorrhizae(SAL) on reverse the multidrug resistance of gastric carcinoma cell line SGC7901/ADR to adriamycin by technologies for proteomics,and then discuss the mechanism of SAL on reverse the multidrug resistance of gastric carcinoma.Methods Immobilized pH gradient-based two-dimentional polyacrylamide gel electrophoresis was used to show the proteins expression in SGC7901/ADR and SGC7901/ADR treated with SAL.The proteins were stained with the gels commassie blue.The differential expression proteins were analyzed by Image Master 2D Melanie 5.0 software.Results In SGC7901/ADR gel and SGC7901/ADR treated with SAL gel,549 and 469 protein spots were identified respectively.There were 87 obviously differential expressional protein spots,36 spots lower and 51 spots higher in quantity in SGC7901/ADR treated with SAL gel.Conclusion The differential proteins in SGC7901/ADR treated with SAL may be related to the mechanism of SAL on reverse the multidrug resistance of gastric carcinoma.
