摘要
目的:建立测定厄贝沙坦氢氯噻嗪分散片中厄贝沙坦和氢氯噻嗪含量测定方法。方法:色谱柱kromasil C_8柱(150 mm×4.6 mm,5μm),流动相为乙腈-10mmol·L^(-1)乙酸胺水溶液(28:72,v/v),检测波长220nm,流速1.0mL·min^(-1),柱温室温。结果:厄贝沙坦浓度在3.0～60.0μg·mL^(-1)(r=0.9998)范围内,氢氯噻嗪浓度在0.25～5.0μg·mL^(-1)(r=0.9995)范围内与峰面积呈良好的线性关系,精密度RSD(n=6)分别为0.2%,0.4%。平均回收率(n=9)分别为101.7%,100.0%。结论:该方法快速简便、准确、专属性强,能有效地控制厄贝沙坦氢氯噻嗪分散片的含量。
Objective：To develop an HPLC method for the determination of irbesartan and hydrochlorothiazide in irbesartan-hydrochlorothiazide dispersible tablets.Methods：The HPLC separation was carried out on a kromasil C8（150 mm×4.6 mm,5μm） column with Acetonitrile-10mmol·L^（-1）ammonium acetate（28：72,v/v） as the mobile phase,detected at 220 nm,flow rate：1.0mL·min^（-1）.Results：The contents of components could be simultaneously determined.The calibration curves were linear in the ranges of 3.0-60.0μg·mL^（-1）（r = 0.9998） for irbesartan,0.25-5.0μg·mL^（-1）（r=0.9995） for hydrochlorothiazide,the RSD（n=6） were 0.2%and 0.4%respectively.The sample was stable in 12h,the average recoveries（n = 9） of irbesartan and hydrochlorothiazide were 101.7%,100.0%respectively.Conclusion：The method is simple,reliable and accurate for the simultaneous determination of irbesartan and hydrochlorothiazide.
