超声强化超临界CO_2萃取复方川芎丹参挥发油成分 被引量:3

Extraction for volatile oil from compound recipe of Ligusticum chuanxiong Hort. and Salvia miltiorrhiza Bge. with ultrasonic enhanced supercritical CO_2

摘要 利用超声强化超临界CO2(USCE)萃取复方川芎丹参挥发油的化学成分,高效液相色谱(HPLC)测定其中阿魏酸、川芎嗪和丹参酮ⅡA的含量,气相色谱-质谱法(GC-MS)分析和鉴定挥发油的化学成分,并和超临界CO2(SCE)萃取法进行对比。正交实验结果得出USCE最佳萃取工艺为萃取压力19MPa、萃取温度35℃、超声电功率600W、萃取时间1h。SCE和USCE挥发油得率分别为2.02%和2.61%。前者挥发油中阿魏酸、川芎嗪和丹参酮ⅡA的含量分别为9.79%、2.49%和0.76%,后者的分别为18.62%、5.79%和0.98%。挥发油经GC-MS检测出的化学成分分别为26种和36种,结果显示USCE法得到的有效成分在种类和含量上都高于SCE法,超声波能有效强化超临界CO2对复方川芎丹参挥发油及其有效成分的萃取。 Ultrasonic enhanced supercritical carbon dioxide(USCE) was used to extract the volatile oil from compound recipe of Ligusticum chuanxiong Hort.and Salvia miltiorrhiza Bge. The contents of ferulic acid,tetramethylpyrazine and tanshinone IIA quantified by high performance liquid chromatography(HPLC) ,gas chromatography mass spectrometry(GC-MS) was applied to identify the volatile oil. The contrast between supercritical carbon dioxide(SCE) and USCE was also implemented. L9(34) orthogonal test table shows the optimized extraction process were:extraction pressure 19 MPa,extraction temperature 35℃,ultrasonic electric power 600 W,extraction time1.0 h. The yield of volatile oil of SCE and USCE was 2.02% and 2.61%,respectively. The content of the compounds contained in former volatile oil were:ferulic acid 9.79%,tetramethylpyrazine 2.49%,and tanshinone IIA 0.76%,and the latter were 18.62%,5.79% and 0.98%. 26 and 36 chemical components were detected in the two kinds of volatile oil,separately.The results demonstrated that ultrasound can effectively intensify the extraction of volatile oil and active ingredients from compound recipe of Ligusticum chuanxiong Hort. and Salvia miltiorrhiza Bge.while applying with SCE.
